We will use the bacterial luciferase (heterodimer of luxA and luxB) venus fusion to replicate a bioluminescence resonance energy transfer (BRET) in E. coli. We are interested in evaluating the use of the improved luciferase in an auto-bioluminescent reporter system. We must first introduce a plasmid that contains the information for the protein called luciferase and luciferin. After the culture is auto-bioluminescent, we can circularly permute the system to achieve a higher yield of light emission.
